Dextran 5 is a biocompatible polysaccharide with low-range molecular weight, offering enhanced stability in biomedical formulations. It is generally widely used in plasma volume expansion, drug delivery, and hydrogel development.

It is a low molecular weight dextran (average Mw ≈5,000 Da) which is a highly purified, non-toxic, branched glucose polymer produced via fermentation of Leuconostoc bacteria. Dextran 5 consists of α-D-glucopyranosyl units with predominantly 1,6 glycosidic linkages and a small percentage of 1,3 branches, resulting in a very water-soluble, white to off-white powder.

Biotechnology: Employed as a stabilizer and cryoprotective agent in protein formulations and cell preservation media.
Research: Serves as a calibration standard in size-exclusion chromatography and as a model polymer for drug delivery studies due to its low viscosity.
Used as an excipient or active ingredient for osmotic regulation in injectables and infusion solutions. It can be a component of plasma volume expander formulations (often in combination with higher Mw fractions).

Handling & Compatibility

Storage: Hygroscopic – store in tightly sealed containers in a cool, dry place to prevent moisture uptake and clumping.
Powder is easy to dissolve; best added to solvent with agitation.
Sterilization: Sterilize solutions by membrane filtration (0.2 µm) rather than autoclaving to avoid Maillard browning or degradation. Molecular weight may decrease slightly if autoclaved. Gamma irradiation of the dry powder can be used as dextran tolerates gamma sterilization well due to its stable glycosidic bonds.
Compatibility: Neutral polymer, so compatible with salts, buffers, and most biomolecules. Does not precipitate proteins and is inert in formulations. Avoid strong oxidizers that may depolymerize polysaccharides and strong acids/bases which may hydrolyze glycosidic bonds over time.

Key Properties

Form: Amorphous powder, white to off-white.
Solubility: >50 g/L in water readily; soluble up to ~25% w/v yielding clear solutions.
Insoluble in most organic solvents (ethanol, acetone, etc.), but soluble in formamide and DMSO.
Viscosity: Very low intrinsic viscosity – even at 10–20% w/v the solution viscosity is modest, which facilitates handling.
Stability: Chemically stable under neutral pH; dry powder is stable for years when stored properly (loss on drying ≤7% per spec).
Resistant to enzymatic breakdown by human amylases (due to branching), and stable over a broad pH range (pH 4–9) in solution.
Available in smaller packs (25-200g) and bulk quantities (> 5kg)
