Dextran 10 is a biocompatible polysaccharide with low to medium-range molecular weight, offering enhanced stability in biomedical formulations. It is generally widely used in plasma volume expansion, drug delivery, and hydrogel development.

It is a low to medium molecular weight dextran (average Mw ≈10,000 Da) which is a highly purified, non-toxic, branched glucose polymer produced via fermentation of Leuconostoc bacteria. Dextran 10 consists of α-D-glucopyranosyl units with predominantly 1,6 glycosidic linkages and a small percentage of 1,3 branches, resulting in a very water-soluble, white to off-white powder.

Biotechnology: Frequently added to cryopreservation media and lyophilization formulations to stabilize proteins and cells during lyophilization. Employed in cell culture as a supplement to enhance osmolality and in nanoparticle coatings to prolong circulation time in research models.

Handling & Compatibility

Storage: Store at room temperature in a dry environment. Dextran 10 is less hygroscopic than very high Mw dextrans, but it will still absorb moisture if exposed – keep container tightly closed. Powder is easy to dissolve; best added to solvent with agitation.
Sterilization: Sterilize solutions by membrane filtration (0.2 µm) rather than autoclaving to avoid Maillard browning or degradation. Molecular weight may decrease slightly if autoclaved. Gamma irradiation of the dry powder can be used as dextran tolerates gamma sterilization well due to its stable glycosidic bonds.
Compatibility: Neutral polymer, so compatible with salts, buffers, and most biomolecules. Does not precipitate proteins and is inert in formulations. Avoid strong oxidizers that may depolymerize polysaccharides and strong acids/bases which may hydrolyze glycosidic bonds over time.

Form: Amorphous powder, white to off-white.
Solubility: Highly water-soluble; 10% solutions are clear and non-viscous, and even up to 30% w/v can dissolve (though viscosity increases with concentration). Insoluble in non-polar solvents; partially soluble in formamide/DMSO if needed.
Viscosity: Low intrinsic viscosity – even at 10–20% w/v the solution viscosity is modest, which facilitates handling.
Stability: Stable as a dry powder (recommended storage <25°C, dry). In solution, it resists hydrolysis at neutral pH and is stable for weeks at 2–8°C.
Resistant to enzymatic breakdown by human amylases (due to branching), and stable over a broad pH range (pH 4–9) in solution.
